Microthesis project:

This microthesis will be oriented to the fabrication and characterization of biochips for the cell
electropermeabilization, for cell transfection or cell fusion purposes. Cell fusion is needed for cancer immunotherapy where dentritic cells have to be fused with cancerous cells. Cell electropermeabilization is also required when artificial gene is expected to be transfected in cells (for instance in the perspective of cell therapy or gene therapy, orsimply for gene reporting). For their electropermeabilization an electrical field has to be applied to living cells,and the efficiency of the operation can be evaluated through fluorescent techniques observation.

  1. The biochip structure we propose to conceive during this microthesis will permit enhance the efficiency of cellelectroporation on a chip, thanks to a control of the field applied to each single cell.
  2. The cells will be captured and arrayed on a microchip using electrical field or/and microfluidic forces. The biochip that will be used for the operation will be fabricated using clean-room microtechnologies.
  3. The student will put in place his experimental benchmark, involving existing microsystems, that he will characterize. He will then propose new technological solutions and/or design , that he will experiment, in order to increase the performances of themethod. Quantification of the efficiency of fusion, or electroporation will be made using fluorescent characterization (fluophore diffusion, FRET, …). Biological characterisation will be made in collaboration with
    biologists.
